Abstract

Carnosic acid and Rosmarinic acid are family of polyphenols that are found in Rosmary plant. They have property biological behaviors such as anti-cancer, anti-viral and anti-oxidants. This study compared the effects of these two compounds based on ceramide metabolism in cell line of Hep- G2. In this experimental study, Hep-G2 cells were cultured in DMEM supplemented containing bovine fetal serum and antibiotics. Cells with double dilution were then cultured from  tow substance Carnosic acid and Rosmarinic acid from concentrations  0 to 70 µM for 24 h and viability of cells was determined by MTT method.  Spectrophotometer was used to measure of caspase-3 activity.To measure the level of ceramide, a recombinant ceramidase acid enzyme and naphthalene-2,3-dialdehyde, which is fluorescent and is connected to sphingosine resulted from ceramidase acid, were added to the cell extract and was ultimately determined by HPLC. In this study also activity of ensymes as sphingomylinase, acid ceramidase, glucosyl ceramide synthase was measured. Carnosic acid increased cell viability dose-dependent in Hep-G2 cells by reducing ceramide levels and decreasing activity of ensymes as caspse-3, sphingomylinase, glucosyl ceramide synthase and  increasing activity of the enzyme acid ceramidase. Rosmarinic acid in concentrations of up to 50 µM decreased cell viability by increasing ceramide levels and activity of caspse-3, sphingomylinase and decreasing activity of  enzymes as acid ceramidese and glucosyl ceramide synthase. Also this substance in concentrations of up to 40 µM caused increasing activity of caspse-3 enzyme. Although in most cases, polyphenols have resulted in induction of apoptosis and decreased cell viability, but in some cases they have inversely affected and caused cell growth.

Abstract

Paclitaxel is a chemotherapy drug inhibiting cell growth. In some studies, patients with normal liver function have experienced increase in bilirubin, ALT and AST by using paclitaxel. The aim of this study was to evaluate the ef-fect of intra-peritoneal injection of doses 5 and 10 mg/kg nZnO on the liver of rats treated with paclitaxel. 35 adult fe-male Wistar rats were divided into 7 groups including control, sham (saline injection), experimental groups1 and 2 (nZnO injection), experimental group 3 (paclitaxel injection), experimental groups 4 and 5 (nZnO and paclitaxel inje-ction). Liver function was examined 28 days after the end of injection. Experimental group3 had large and swollen liver morphology. Most hepatocytes had dense nuclei and changed cell shape indicating of cell death. Blood test showed si-gnificant increase in the levels of ALT, AST and bilirubin and decrease in the level of ALP in comparison with the co-ntrol group. In experimental groups 4 and 5, cell shape alterations, increase in cell death and increase in liver markers were remarkably reduced in comparison with the experimental group 3, in a way that there were no significant differ-ences with the control group. No significant differences were observed between the control group and experimental gr-oups 1 and 2. According to the findings, nZnO can reduce the side effects of paclitaxel on liver tissue.

Abstract

Cutin is a polymer that is constructed in plants by the condensation and oxidation of fatty acids and plays a key role in the protection of plants against pathogens. Cutinase is a hydrolase enzyme that breaks down the cutin. The purpose of this study was to extract cutin from red apples with oxalate buffer, cutinase enzyme activity assay in LB culture, and bioinformatic analysis. To attain these purposes the cutinase-producing strains that had previously been isolated were inoculated in culture medium containing cutin. After initial culture, the bacteria were cultured in LB medium and cutinase activity was measured using the p-Nitrophenyl butyrate. In order to execute bioinformatic analysis, the isolated sequences of six cutinase-producing bacteria were analyzed based on computational data bases and their phylogenetic trees were prepared. Then, the similarities in the sequences of a large number of cutinase-producing samples were analyzed by drawing the phylogenetic tree. The results showed the separation of cutinase-producing prokaryotes from cutinase-producing eukaryotes. Then, the sequence of 16S rDNA of these cutinase-producing samples as well as the samples we had prepared were evaluated and their phylogenetic relationships were determined. This analysis showed that the new sequence stood alongside the bacterial samples. Thus, our cutinases may be similar with these bacterial cutinases in structure and function.

Abstract


To investigate the effect of herbicide Trifluralin and the role of magnetized water on enzymatic activities and content of proline and growth substances including gibberellin (GA3) and cytokinin (6- benzyl amino purine), an experiment was conducted using squash (Cucurbita pepo L. var. Shiraz Hybrid F1) seedlings. Four levels of trifluralin (0, 5, 15 and 25 ppm) and irrigation by distilled and magnetized water were applied in a completely randomized design in 3 replicates at Urmia University in 2016. Different levels of trifluralin were added to pots 2 days after planting. Seedlings were treated by modified Hoagland solution with diurnal temperature 30:18 °C, relative humidity of 70-80 percent and light period of 16:8 hr, during a 5-week growing period in growth chamber. The effect of magnetized water and trifluralin on ornithine amino-transferase (OAT), proline dehydrogenase (PDH) and content of proline in both shoots and roots as well as GA3 and cytokinin was significant (p>0.01) according to ANOVA. The content of proline and OAT in roots and shoots as well as GA3 and cytokinin increased significantly, although the PDH of roots and shoots decreased under the influence of magnetized water. The content of proline and OAT increased and GA3, cytokinin and the activity of PDH declined by the increase of the levels of trifluralin. Overall, it can be concluded that the growth and biochemical indices of squash plants were improved by magnetized water under toxicity of herbicide trifluralin.

Abstract

Nowadays plant endophytic bacteria have found diverse and useful applications in biotechnology; therefore, much attention has been paid to the isolation, identification, and evaluation of these microorganisms. Since the sterilizing plant tissue surfaces from epiphytic bacteria is difficulty, the efficacy of three different screening methods for endophytic bacteria including 1- HClO sterilization, 2- Periodic sterilization (modified tyndallization) and 3- Triton X100 and HClO sterilization, was evaluated in this study. The modified Tyndallization is an innovative method used in this study to appropriately remove the internal spores of epiphytic bacteria, considered to be an obstacle to the isolation of endophytes. Most of the endophytic bacteria were isolated from dicotyledons and leaves. Endophytic bacteria were also studied for the production of different hydrolase enzymes, whereas the protease enzyme was produced in a wide range of endophytic bacteria in greater quantities than other enzymes. The EndoA strain was molecularly identified and found to be 100% similar to Bacillus halotolerans.

Abstract

Comparison of morphological structures is a key element in evolutionary biology and taxonomy, as well as in the studies concerning variation and biodiversity either at interspecific or intraspecific levels. This study aimed to compare the bony structures of Chondrostoma orientale and C. nasus. For this purpose, three specimens of C. orientale from the Kor River (passing through Iran), and five specimens of C. nasus from the Danube (passing through Germany, Austria, Slovakia, Hungary, Croatia, Serbia, Romania, Bulgaria, Moldova and Ukraine), Dnieper (passing through Russia, Belarus and Ukraine) and Rhône river basins (passing through Switzerland and France), as well as Lake Ohrid (Located between Macedonia and Albania) were analysed. The results showed that the two anterior lateral processes of supraethmoid bone were more distant in C. orientale than those in C. nasus. The anterior profile of the supraetmoid bone was concave in C. orientale, while it was almost flat in C. nasus. The posterior pharyngeal process in the basioccipital was wide and triangular in C. orientale and narrow and rectangular in C. nasus. The maxillary mid-lateral ascending process was slightly inclined forward in C. orientale, while it was slightly inclined backward in C. nasus. The maxillary distal process was long in C. orientale and it was short in C. nasus. The premaxillary had a rostral ascending process in C. orientale, while it was missing in C. nasus. The anterior portion of the dentary was narrow and elongated in C. orientale and the coronoid process was perpendicular and located in the middle of the dentary; in contrast, the anterior portion of the dentary was short and wide in C. nasus and the coronoid process was inclined forward.
