Abstract

Mesquite is an invasive and problematic weed in warm and dry areas and so its management is very important. In order to study the effective factors on mesquite seed germination, the following studies were conducted to investigate: 1- seed viability; 2- the effect of constant temperatures on seed germination including 5, 10, 15, 20, 25, 30, 35 and 40°C independent dark and independent light conditions; 3- Effect of temperature fluctuations on seed germination including 10/0, 20/10 and 30/20°C; 4- the effect of scarification with concentrated sulphuric acid and physiacal scarification on seed germination; 5- the effect of planting depth seed on seedling emergence; 6- the effect of drought stress including 0, 100, 200, 300, 400, 500, and 600 Mm PEG on seed germination; and finally 7- the effect of salt stress including 0, 100, 200, 300, 400, 500, 600, and 700 mM NaCl on seed germination. All experiments were performed as completely randomized designs with 4 replications. The results showed that seed viability of mesquite was 81%. The optimum temperature for seed germination was a constant temperature- 30°C. In general, constant temperature was more suitable than temperature fluctuations for seed germination. The best seed planting depth for seedling emergence was 2 cm. The most seed germination was observed under the effect of 20 min scarification with concentrated sulphuric acid. Physical scarification had no significant effect on seed germination. While the study of salt and drought stresses effect showed that the most seed germination (after control without NaCl and PEG), was due to concentrations of 100 and 200 mM, seed germination decreased with the increase of NaCl and PEG concentrations. Based on the present results, light did not play a crucial role on seed germination. Therefore, mesquite seeds were not photoblastic. These characteristics are very important in making mesquite an invasive weed. Having precise information of these traits, enables us to have a better control over the management of this troublesome weed.

Abstract

Recently, concern over environmental and health effects of massive use of antibiotics has led to the growth of bio-control agent application. Poly-β-hydroxybutyrate (PHB) is a natural polymer that can be depolymerized into water-soluble short-chain fatty acid monomers and acts as a microbial control agent. In this study, the effects of the addition of PHB to the diet of Siberian sturgeon fingerlings by 2% and 5% were investigated. Community Level Physiological Profile (CLPP) was used to analyze anaerobic bacterial metabolic diversity in Siberian sturgeon fingerlings hindgut by using Biolog™ Ecoplate microplates. The results indicated that PHB increased metabolic activity in anaerobic bacteria in sturgeon hindgut. Also Lorenz curve and the Shannon index of Biolog™ Ecoplate data revealed that anaerobic metabolic potential of the bacterial community was different in the PHB-treated fishes compared with the control situation.

Abstract

The aim of this study was to assess the effect of the smoke of Verbascum speciocum leaves grown in 'Bouein Zahra' region, on the healing of skin lesions in Wistar rats. In this study in vivo conditions, 36 adult male Wistar rats were randomly divided into 3 groups. The operation day was considered as day zero and the procedure took 10 days. After the end of the treatment, i.e., on the 14th day, rats were killed by choloroform and samples were collected for histological analysis Masson trichrome and Vangason staining for collogen and hydroxyprolline amino acid OD assessment was applied. Studies in experimental groups exposed to the smoke of the leaves revealed that there was no increase in epidermis thickness, hair follicles diameter, skin, nor in the number and diameter of blood vessels as compared with control and sham groups. However, the amount of hair follicles was decreased. There was an increase in collogen diameter but this increase was not significant. But a significant increase was observed in OD of hydroxyl proline amino acid assessment (p<0.001) in experimental groups compared with sham and control groups. The results indicated that the use of smoke of V.speciocum leaves on the skin lesion had absolutely no increase on the wound healing ratios and its closure; it maximally could had very little effect on the proliferation as one step in the repair process, which needs further investigation.

Abstract

The tubers of Bongardia chrysogonum, belonging to to Podophyllaceae, are rich in alkaloids. In this study, total alkaloid content of the tubers of this plant in Naghadeh, Zanjan and Boukan were determined by spectrophotometeric method. In addition, the effects of soil characteristics such as total nitrogen, potassium, pH, soil texture and tubers nitrate levels on the plant total alkaloid content were measured. The results of the analyses indicated significant differences between the total alkaloid content of B.chrysogonum in these three regions. Moreover, the results showed significant differences among nitrates amounts of this plant in these habitats. The highest and lowest nitrate contents belong to Naghadeh and Boukan plants, respectively. The evaluation of soil samples of these three regions indicated that the texture of soil in Naghadeh is clay-loamy, in Zanjan it is silty- clay- loamy and in Boukan it is loamy- sandy. It was also discovered that increase in plant total alkaloid content depends on the increase of sand percentag in soil to some extent. The effect of soil pH on total alkaloid content turned out to be non-significant.

Abstract

This investigation was carried out to evaluate Indole acetic acid (IAA) production by isolated bacteria from drought and salinity affected soils. Four hundred bacteria were isolated from soils with different levels of electrical conductivity (EC) and sodium adsorption ratio (SAR). Then the ability of these bacteria to produce IAA and the effects of different amounts of tryptophan and drought-salinity tensions on IAA production were analyzed. Results indicated that the effects of drought and salinity tensions on variability of IAA production were statistically significant. One of the bacterial species isolated from saline-sodic soils, Arthrobacter siccitolerans, was IAA producer and data comparison indicated that under drought stress conditions it produced IAA 84.3% higher than isolate RS11. While isolated RS11 produced highest rate of IAA (10.42 µg.ml-1) under normal conditions. Also, A.siccitolerans continued to produce IAA until the EC was 40 dS.m-1 and OP (Osmosis pressure) was -20 bar. The amount of IAA production per unit of applied tryptophan in this bacterium was 0.1 and in others it was lesser. According to experiments, the relative preference of this bacterium was verified and proposed for field experiments in drought, salinity and salinity-sodicity affected soils.

Abstract

In this study, water stress was applied by polyethylene glycol at a concentration of 20 perecentage. To improve the resistance of the plants, the samples were treated by potassium nitrate at concentrations 5, 10, and 15 mM within 9 days. Changes in proline, total protein, photosynthetic pigments, carotene, anthocyanin, malondialdehyde, phenols, flavonols, flavonoids, soluble sugars and potassium ion were examined. The results showed that tobacco plants which had been exposed to drought used the accumulation of osmolytes such as proline, soluble sugars and potassium in order to balance their osmotic pressure. Drought stress also caused oxidative stress and increased the production of active forms of oxygen. As a result, non-enzymatic antioxidant defense system of tobacco plants including anthocyanins, flavonoids, flavonols and beta-carotene increased, which could be considered to be a major step for resistance to drought. The results also showed that the concentration of 15 mM potassium nitrate in particular, could significantly improve some of the harmful effects of stress and reduced photosynthetic pigments and proteins. Potassium nitrate could also bring down the MDA and beta-carotene levels to equivalent levels in control plants. As a result, it seems that using potassium can affect plant resistance to drought and plays an important role to reduce some harmful effects of stress.

Abstract

It is clear that DNA concentration and biological detection devices have many applications in the fields of genetics and biotechnology. For this goal, some similar devices have been designed working with the laser light which the disadvantages of them are high prices and the use of significant amounts of sample volume. But this designed device which works with LED light has very cheap manufacturing cost. The amounts of DNA used in this device are very low, about a few micro-liters and measurement of small, fast and portable it is also the advantages of these devices. The Data recorded as voltage versus time and the Fourier transform the frequency domain. From the frequency curve, by using the Byrlambrt it is possible to obtain the relative concentrations of DNA, viruses and assays measuring DNA damage and many other applications. By use of this device, the concentration of DNA colored with three different pigments was measured.

Abstract

Basolateral Amygdala is an important site of anxiety. Interactions between α-adrenergic and opioidergic systems in Basolateral amygdale were used for investigation anxiety and memory. The elevated plus-maze has been employed. The male wistar rats were used for this test. The site of BLA was cannulated bilaterally. Rats we injected morphine (4, 5.6 mg/kg) intraperitonealy, while clonidine (1, 2, 4 µg/rat) and yohimbin (0.5, 1,2 µg/rat) were injected to BLA. Open arm time percentage (%OAT), open arm entry (%0AE) and locomotor activity were determined by this behavioral test. Retention tested 24 hours later. Administration of morphine (6mg/kg) increased the OAT% in anxiety test, indicating anxiolytic-like effect. Intra Basolateral amygdala infusion of clonidine (4μg/rat) has an anxiolytic-like effect. While co-administration clonidine (4μg/rat) and in effective dose of morphine (4mg/kg) showed significant increase of OAT% in anxiety test; thus presenting anxiolytic response. Intra Basolateral amygdala administration of yohimbine (2μg/rat) decreased OAT% indicating of decrease anxiety-like behavior. While co-administration of intra Basolateral amygdala clonidine (4μg/rat) and effective dose of morphine (6mg/kg) showed a significant increase of OAT%, presenting anxiolytic response; co-administration of ineffective doses of morphine (4mg/kg) and yohimbine (1μg/rat) with the effective dose of clonidine (4μg/rat) showed that yohimbine could reverse the anxiolytic-like effect of morphine and clonidine. It should be noted that there are no significant changes in locomotor activity. The results indicate that morphine creates the compromise changes in adrenergic neurons of Basolateral amygdala by changing the α-adrenergic system on anxiety.

Abstract

Lipase is one of the most important hydrolytic enzymes widely used in various commercial activities such as food, dairy, pharmaceutical and detergent inducteries. In this experiment, Taguchi method was attempted as a powerful method to optimize the factors affecting enzyme production and to investigate the interactions among these factors and their optimum combination in Salinivibrio sp. SA2. The optimum conditions for pH, temperature, shaker's rpm, olive oil concentration and salt type turned out to be 8, 35 °C, 100 rpm, 2% and sodium chloride 1 M, respectively. Significant factors influencing on the lipase production proved to be pH, agitation and Salt type. The maximum lipase activity in optimum condition and at the 5% significance level (p< 0.05) was 120.4 U/mg.

Abstract

Septohippocampal system plays an important role in regulating fear and anxiety behaviors. In this study, the effects of histamine injected into the dorsal hippocampus and opioidergic agents into medial septum on the anxiety-like behaviors in rats were analyzed, using the Elevated Plus-Maze (EPM) test. Injection of 1 and 5 μg/rat histamine into dorsal hippocampus had no effect on anxiety-like behavior, while injection of 10 μg/rat histamine increased the percentage of open arm time (%OAT) and open arm entry (%OAE), which indicated the anxiolytic effects of histamine. Microinjection of morphine, μ-opioid receptor agonist, into the medial septum (1μg/rat) increased the (%OAT) and (%OAE). Doses of 0.25, 0.5 μg/rat morphine had no effect on anxiety. Co-administration of histamine ineffective dose (1μg/rat) to the dorsal hippocampus and ineffective dose of morphine (0.25µg/rat) to the medial septum increased the (%OAT) and (%OAE). Subsequently, injection of different doses of naloxone (1, 2, 4 µg/rat), as an opioid receptor antagonist, into the medial septum in the presence and absence of an effective dose of histamine (10 µg/rat) in the dorsal hippocampus, was studied. Injection of naloxone (4 µg/rat) into medial septum decreased the (%OAT) and (%OAE), but did not alter the locomotor activity, which indicated the anxiogenic effects of naloxone. Simultaneous injection of histamine (10 µg/rat) into dorsal hippocampus with doses of naloxone (1, 2, 4 µg/rat) into the medial septum, indicate anxiolytic effects and increased %OAT and %OAE in Elevated Plus Maze, although when the dose of naloxone was 4μg/rat, this effect was less observed. The results indicate that hippocampus histaminergic system interact with medial septum opioidergic system and the interaction of these systems modulates anxiety behavior.

Abstract

This study aimed to recycle sawdust and blood powder using Eisenia foetida earthworm and investigate some biochemical changes of these waste materials during vermicomposting. Blood powder was mixed with sawdust in proportions of 0, 5 and 10% and the mixture was allowed to pass through earthworm guts for four months. At intervals of 1, 2, 3, 4, 6, 8, 12 and 16 weeks, the biological activity (i.e. basal respiration), pH, EC, total organic carbon, total nitrogen and C:N ratio were determined. The results showed that the period of time, the concentration of blood powder and the interaction between these two significantly affected all parameters. As the incubation time increases, the cumulative amount of mineralized carbon, the total nitrogen and EC increase in all vermicompost treatments whereas organic carbon, C:N ratio and  pH decreased. Adding blood powder to sawdust bed resulted in an increase in carbon mineralization rate, EC and total nitrogen while pH, organic carbon and C: N ratio decreased. At the end of incubation time, the treatment of sawdust with 10 % blood powder resulted in the highest amount of released CO2 (142.1 µg C g-1), EC (3.7 dS m-1) and total nitrogen (2.24 %) and the lowest amount of pH (6.6), organic carbon (22.5 %) and C: N ratio (12.4). According to the results, the process of vermicomposting can be used as a safe method for the disposal of sawdust and blood powder

Abstract

This study aimed to investigate the protective effect of Arctium lappa (AL) on gentamicin (GM)-induced nephrotoxicity in rats. Twenty-four Wistar rats were divided into four groups including: control group; GM group (intrapritoneal injection, IP, of 100 mg/kg GM B.W.); GM+AL group (received IP injection of 100 mg/kg GM and 500 mg/kg AL orally) and AL group (received 500 mg/kg AL orally). The experimental period lasted for 10 days. Nephrotoxicity was biochemically and histologically evaluated. The concentrations of creatinine, urea, malondialdehyde (MDA), superoxide dismutase (SOD) and peroxide hydrogen (H2O2) in the serum samples were determined. Moreover, histological examinations were performed. The animals treated with gentamicin showed significantly higher serum urea, creatinine, MDA and H2O2 levels and lower SOD activity. However, co-administration of AL produced amelioration in biochemical indices of nephrotoxicity in serum. Histomorphological examination showed necrosis and desquamation of tubular epithelial cells in the renal cortex in animals treated with gentamicin whereas simultaneous administration of AL and GM reduced histological damages. The data obtained suggest that treatment with AL extract can help to reduce gentamicin-induced nephrotoxicity.

Abstract

Special properties and various applications of Spirulina species have led to the growing focus of biotechnology to the cost effective cultivation. Open ponds used in the most commercial cultivation have many deficiencies such as risk of contamination, lack of light penetration, inefficient mixing and low productivity. In this study, an innovative reactor was designed and manufactured. By adding a control area, it was possible to control some environmental conditions for a limited amount of open pond volume. The conventional pumping system was replaced by a new elevator system to reduce the damage of to the fragile cells. The baffles structure and aeration from bottom, contributed to better mixing system. The effect of tree factors (Circulation Cycle (Cir. Cyc.), Control Volume (Con.Vol.) and Irradiance Intensity (Irr. Int.) in units of hours, percentage and Lux respectively) on the quantity of biomass (Specific Growth Rate (SGR, day-1) and fifth and seventh day dry weight (g/L)) were evaluated using RSM-CCD. The maximum biomass dry weight (0.915 g/L) was achieved in longest circulation cycle (12 hours), 30% Con. Vol. and 7000 Lux light intensity. The optimal irradiance intensity to achieve the highest biomass was 8700 Lux. According to the results, due to the great influence of Con. Vol. and Cir. Cyc., it is possible to increase the productivity and enhance the culture dry weight by controlling a limited amount of ponds volume (20%), subject to economic considerations. These successful results can provide the context using a control zone for several open ponds to improve the overall efficiency.

Abstract

In order to investigate different treatment effects on germination in Gundelia tournefortii, three factorial experiments were carried out in the completely randomized design with three replications. The first factor was stratification at 4oC in three levels including 0, 3 and 6 weeks, the second factor was mechanical scarification in two levels including seeds with/without scarification and the third factor was chemical treatments of seed dormancy breaking in three levels including distilled water and gibberellic acid at concentrations of 1000 and 1500 mg/l and Thiourea 0.1%. The results showed that the effects of stratification, scarification and chemical treatments of dormancy breaking and their interactions were significant on all studied parameters (p<0.01). Mean comparison values were significant for the effects of spate treatments together with mechanical scarification for the improvement of all traits. The results also revealed that treatment of mechanical scarification treatment together with gibberellic acid 1000 mg/l in six weeks stratification, due to maximum germination speed, seedling dry weight, seedling length, large-scale germination percentage and seedling vigor index and modest average germination period, was the most influential treatment for seed dormancy breaking of this plant. Correlation between germination percentage with germination rate, dry weight and seedling length was significant. It seems that seed dormancy type in Gundelia tournefortii is physiological and physical, as eliminating physiological and physical factors leads to its germination improvement.

Abstract

The Basolateral Amygdala (BLA) has modulatory effects on working and reference memory. The aim of this study was the evaluation of effects of dopaminergic system in the BLA of rats on working and reference memory behaviors. The number of working and reference errors and time spent in the arms by rats were measured in the radial arm maze according to DSWS protocol. The animals were cannulated in the BLA bilaterally. The microinjection of low dose (0.005µg/rat) and high dose (0.5 µg/rat) of apomorphine have indicated a significant decrease in number of working memory error. But, there was not any change in the numberof reference memory error that showing the improvement of working memory. While the injection of moderate dose apomorphine (0.05 µg/rat) increased these parameters and also enhanced the spent time in working arm that presenting both memories damage. The chlorpromazine injection (2 µg/rat) decreased the number of working and reference memory errors representing the improvement of these memories. Microinjection of chlorpromazine (2 µg/rat) with different doses of apomorphine had no significant change on the both number of errors and the time spent in comparison with control groups. These findings show that BLA dopaminergic system modulates the working and reference memory through that both of (D1/D2) receptors. Also, the effect of this system in BLA is the resultant function of the both receptor families.

Abstract

11, 25 dihydroxy vitamin D3, an active metabolite of vitamin­D3 has been reported to inhibit the growth of number neoplasms such as prostate, breast, colorectal, leukemia and skin cancers. Valproic acid, as a potent histone deacetylase inhibitor, also plays an important role in inhibition of proliferation of tumor cells. However, there are no reports so far on the cooperation between valproic acid and vitamin­ D3 for anti-leukemic effect. The goal of the present research was to evaluate whether low doses of vitamin D3 potentiate the toxicity of valproic acid and whether this toxic action is mediated via apoptotic mechanisms. In this study HL-60 cells were treated either with different concentrations of valproic acid and vitamin­ D3 alone and in combination with each other for 24 hours. Cell survival was determined by MTT assay and then Hoechst staining was used to determine the type of death cell. This present study indicates that vitamin ­D3 potentiates the antitumor effects of valproic acid. Also, the results of staining cells showed that valproic acid and vitamin ­D3 induced apoptosis in HL-60 cells. In total, the new combination of valproic acid and vitamin D3 showed synergistic anti-proliferative effect and induced apoptosis on HL-60 cancer cells.

Abstract

Feeding frequency and stocking density are important factors affecting growth and maturation of cultured fishes. The aims of this investigation were to determine the effects of stocking density (10, 20, 30 and 40 fish per aquarium) and feeding frequency (2, 3 and 4 times per day) on growth parameters and hematocrit of the Rutilus rutilus caspicus. The experiment was continued for 45 days in two replicates. The results of this study showed that growth parameters, including weigh gain (WG), feed conversion ratio (FCR) and specific growth rate (SGR) were affected significantly by stocking density (P<0.05), but not significantly affected by feeding frequency (P>0.05). The highest WG and SGR were recorded with the lowest stocking density (10 fish / aquarium). The least FCR was obtained in density of 10 fish/aquarium, FCR value was significantly higher with high stocking density (P<0.05). Also, interactions between stocking density and feeding frequency were no significant on growth parameters (P>0.05). Density and feeding frequency hand no significant effect of hematocrit concentration (P>0.05). On the other hand, interactions between stocking density and feeding frequency significantly affected hematocrit levels (P<0.05). The lowest level of hematocrit was observed in density of 10 fish/aquarium at two times / day feeding frequency. The results of this study indicated that density is significantly affected the growth, but increasing of feeding frequency there is not effect on growth in the Rutilus rutilus caspicus.

Abstract

New properties of nano-materials have made nanotechnology the leading part of biology and medical sciences. Due to their various biomedical properties, iron-based magnetic nanoparticles (MNPs) have been highly considered by biological researchers. Nowadays, increasing resistance to antibiotics is a major problem in treating clinical infections. Finding new antibacterial agents is therefore essential for the treatment of resistant strains. In this study, the iron oxide MNPs were produced using culture-medium supernatant of a newly isolated bacterium to investigate the inhibitory effects of the NPs on strains with a major role in clinical infections. Biosynthesis of iron oxide MNPs were detected by UV-Vis spectroscopy and the average size of particles was estimated by dynamic light scattering technique. The anti-bacterial activity of these NPs against E. coli and S. aureus was investigated using methods for the calculation of bacterial sensitivity coefficient. In the presence of NPs, the highest sensitivity coefficient value was observed for E. coli in 1xMIC concentration. On the other hand, S. aureus showed the lowest value. The death rate of the two strains in contact with NPs followed the first order kinetic equation and the survival rate decreased with the increase of exposure time. The results of this study as well as the high functionality of iron oxide MNPs, make its application desirable in the prevention and treatment of clinical infections.

Abstract

In this study, the cytotoxic effects of aqueous and ethanolic extracts of Sedum album L. on human stomach cancer cell line (AGS) and breast cancer cell line (MCF-7) were evaluated by MTT, BrdU and TUNEL assays. The results demonstrated that both extracts had antiproliferative and apoptotic effects in a dose-dependent manner. The MTT assay data revealed that the AGS cell underwent more cytotoxicity in comparison with the MCF-7 cell. It also revealed that ethanolic extract was more potent than aqueous extract. The BrdU assay results showed that the proliferation of AGS and MCF-7 cells was reduced to 50% and 43%, respectively, at the highest concentration of the aqueous extract. In addition, the ethanolic extract reduced the proliferation of AGS and MCF-7 cells to 75% and 60%, respectively. The AGS and MCF-7 cells underwent 52% and 12% apoptotic death upon treatment by the ethanolic extract as TUNEL assay showed. The aqueous extract induced 28% and 25% apoptosis in the AGS and MCF-7 cells, respectively. Both inhibition of proliferation and induction of apoptosis are desirable strategies for cancer treatment among researchers. Identification of S. album compounds and analyzing their effects in animal model of cancer can help us with understanding its anti-cancer properties.

Abstract

The identification of the characteristics of native halophytic species is very important for their practical application. The present study was carried out to identify the halophytic species of a desert and highly saline region around the city of Khosf, southern Khorasan Province, and to detect their general tolerance mechanisms (i.e. salt exclusion or inclusion). Sodium accumulation in the roots and shoots of seven species, including Launaea arborescens, Peganum harmala, Pteropyrum olivieri, Artemisia santolina, Zygophyllum eurypterum Boiss, Aerva javanica, Pulicaria gnaphalode, and their rhizosphere soil were determined. The bioconcentration of sodium from soil to roots (BCF), its translocation from roots to shoots (TF), and its accumulation in the shoots (AF) were then calculated. Results showed that the soil of the studied area was clay loam with a high EC of 65 ds/m, indicating its high salinity level. According to the values of sodium adsorption ratio (SAR) and the exchangeable sodium percentage (ESP) and regarding EC, the soil of studied region can be regarded as a saline-sodic soil. The value of TF in some species was higher than 1. However, none of the species had BCF and AF greater than 1, thus, none of them could be considered to be salt accumulator. Instead, it could be assumed that all of the studied species were salt, or at least, sodium excluders.
