Abstract

Cholestasis caused by the excessive accumulation of bile within the liver, due to intrinsic or extrinsic factors. Cholestasis side effects are associated directly or indirectly with the reduction of bile flow and the confinement of materials related to bile secretion (such as bile acids, bilirubin, and cholesterol). On the other hand, some factors such as opioids, alkaline phosphatase, endotoxin and nitric oxide increase in blood, which could cause tissue damage. Since water intake reduces during cholestasis and hypothalamic nuclei such as paraventricular and supraoptic nucleuses are involved in the regulation of body water; Therefore, in this study, the histopathological changes of hypothalamic nuclei were evaluated. Male Wistar rats weighing 200–250 g were randomly divided into three groups. Three sets of seven groups were unoperated control, sham-operated and bile duct-ligated rats. The tissue samples were analyzed using histotechnique and light microscope. Brain tissue necrosis in paraventricular and supraoptic nucleus in cholestatic rats increased, but in the sham and control rats no changes were observed and also cholestasis caused wrinkle chromatic nuclei and increased thickness of hypothalamic nuclei. Because endotoxin causes tissue trauma, it is likely increased endotoxin may leads to tissue changes in the brain.

Abstract

Cotton (Gossypium hirsutum L.) is an important crop in the world and increasing its fiber quality is very cr-ucial for textile industries. Spider silk is the strongest and most elastic fiber ever known in the nature. Cotton is one of the main crops in the world and increasing its fiber quality is very important for textile industries. In this regard, a synt-hetic construct has been designed to offer spider silk quality to cotton fibers by fiber-specific expression of Major am-pullate spidroin1 (MaSp1) gene under control of core sequence of GaRD22-like1 promoter. The synthetic construct was double digested by the EcoRI and NheI and sub-cloned in pCAMBIA1304 binary vector. E.coli DH5α was transformed using new plasmid namely pCSP. Ligation and intact backbone of plasmid was conformed using MaSp1 and hygrom-ycin re-sistance genes specific primers and finally with EcoRI/NheI double digestion. Agrobacterium tumefaciens LBA4404 was transformed with pCSP to transform cotton ovules of Varamin cultivar. The expression of MaSp1 in co-tton ovules and fibers showed that this synthetic sequence had been successfully expressed under control of GaRDL1 core promoter and this construct without codon optimization could be used for cotton transformation and its fiber qual-ity manipulation.

Abstract

In cholestasis syndrome, the change of bile acids, salts, endotoxins and opioidsis is accompanied with hepaticencephalopathy and brain trauma; therefore, in this study, the histopathological changes of hippocampus after bile duct ligation were investigated. This study was performed on male Wistar rats. The rats were divided into three groups, namely the control, sham and cholestatic groups. After two weeks, the rats were killed under anesth-esia and their brains were dissected with no delay. The specimens were processed routinely and were sectioned into slices of 6-micron thickness. The sections were stained by Hematoxiline-Eosin (H&E) method. Subsequently, they were studied using optical microscope. Necrotic cells, the diminution of cell density and wrinkled chromatic nuclei were observed in hippocampus section. In addition, the thickness of hippocampus was mostly decreased. However, in the sham and control groups no changes were observed. Since endotoxin causes tissue trauma, the increase of en-dotoxin may result in tissue changes in the brain.

Abstract

Purslane (Portulaca oleracea L.) contains valuable secondary metabolites such as Dopamin, Noradrenaline and Omega-3. This plant is used in various medicinal, food and hygienic industries as well as the treatment of different diseases such as diabetes, heart disease and pain relief. Callus induced from medicinal plants are used to increase the production of secondary metabolities in cell suspension culture and gene transfer. The purpose of this experiment was the study of different concentrations of BAP and 2,4-D of two explants from leaf and shoot tips to produce callus. Leaf and shoot tip explants were used in MS with different concentrations of BAP at three levels (0, 1 and 2 mg/L) with 2,4-D at three levels (0, 0.5 and 1.5 mg/L). Results showed that interactions between hormones and explants were significant in the percentage of callus induction, fresh weight and callus diameter at 1% level. The best result which was the leaf explant with 100% callus induction, 121 mg fresh weight and 5.106 mm callus diameter was obtained by the combination of BAP 2 mg/L and 2,4-D 0.5 mg/L. Shoot tip explants with 75% callus induction, 106 mg fresh weight and 3.03 mm diameter was obtained by the application of 1 mg/L BAP and 0.5 mg/L 2,4-D.

Abstract

Aloe vera L. is one of the most valuable plants in the pharmaceutical, cosmetic, sanitary and food industries. In vitro culture is used for commercial production and due to the abundant application of this plant, extensive research has been performed on the in vitro culture of Aloe vera. For this purpose, the present study was conducted at two stages. At the first stage, the best method of sterilization of explants derived from Aloe vera offshoots was investigated. At the second stage, the effect of the type of explants, the light condition (dark and bright) and the effect of BAP (Benzyl Amino Purine) and NAA (α-Naphthalene acetic acid)) growth regulators on regeneration and the amount of phenolic compounds were studied. A factorial experiment was executed on the basis of a completely randomized design with three replications. The best sterilization protocol was 0.1% mercuric chloride (for 2 minutes), 70% ethanol (for 30 seconds) and 15% sodium hypochlorite (for 5 minutes). The little white explant derived from the base of leaves, with the lowest percentage of phenol and the highest survival rate (67.5%) in darkness, was found to be the best candidate. MS medium supplemented with 0.75 mg / L BAP and 0.25 mg / L NAA resulted in the highest stem number (2.5) and stem length (42.107 mm), establishment percentage (73%), leaf number (6.33), leaf diameter (4.8 mm), chlorophyll b (9.216 mg/g) and carotenoids (4.81 mg/g). The highest content of chlorophyll a (56.07 mg/g) and total chlorophyll (61.35 mg/g) were found in samples treated with hormonal medium, supplemented with 1.5 mg / L of BAP with 0.5 mg / L of NAA. The maximum number (3) and average length (33.3 mm) of roots were observed in samples treated with the hormone-free medium.

Abstract

High-quality DNA extraction plays an important role to make sharp bands in the gel electrophoresis and also produces clean chromatograms. Usually, DNA extract is delivered using the modified CTAB method but this method cannot obtain high-quality DNA for molecular analysis from old dried leaves of Bellevalia due to having different chemical compounds which inhibit to obtain a clear DNA extraction. To solve the problem, in different phases of a modified CTAB protocol, activated carbon, phenol, potassium acetate, RNase and deionized water (instead of TE) were applied to eliminate contaminants and improve obtained high-quality DNA. The DNA absorption data from the modified method were compared with CTAB method in 260 and 280 nm wavelengths. The results showed a 15-80-fold change in the DNA concentration. This study suggests the modified method of extraction is more efficient in the quality of DNA obtained from the little amount of herbarium old leaves of Bellevalia than that of the CTAB method.

Abstract

It has been found that the second isoform of COX enzyme known as COX-2 plays an important role in inflammation and rheumatoid arthritis and osteoarthritis. Thus, designing COX-2 inhibitors to treat inflammation is among the most important goals of researchers. In this study, the inhibitory effect of 3 new imidazole derivatives on COX-2 was evaluated by in silico approach. Molecular docking was done using Autodock Vina and the best binding mode of inhibitors was used as input of molecular dynamics (MD) simulation. MD was performed using Gromacs software for 120 ns. Then, structural and thermodynamic analyzes (ΔGbinding) and prediction of physicochemical properties were performed. RMSD data showed the compounds reached a good equilibrium and had favorable stability during simulation. Also, the RMSF showed that due to binding of inhibitors, the fluctuations of complexes decreased and the active site residues had the lowest amount. Rg, SASA and DSSP analysis showed that the protein structure did not change significantly. It was also found that Ser530 and Tyr355 residues play a more effective role in hydrogen bond formation. Physicochemical parameters determined the good drug-likeness properties for all compounds. Structural and thermodynamic analyzes (MM-PBSA) and IC50 data indicate the favorable inhibitory effect of compound 5b.

Abstract

Legumes are common crops in arid and semi-arid regions. This study was conducted to investigate three levels of salinity stress on 18 lentil cultivars in the greenhouse and laboratories of Mohaghegh Ardabili University in 2016-2016 in a factorial manner in the form of a randomized complete block design with three replications. The results of comparing average lentil genotypes were done with Duncan's method at the five percent probability level. Correlation was done in separate stress levels. The dendrogram of cluster analysis divided the genotypes into three groups in control and 60 mM conditions and two in 120 mM conditions. Genotypes 7 and 5 were in the top group in all groupings. The relationship between ISSR molecular markers and physiological traits was calculated among genotypes. All traits in three levels of salinity stress had a significant correlation with some ISSR markers. A total of 22 positive markers for physiological traits were identified at the control level, 9 positive markers at the moderate stress level, and 23 positive markers at the severe stress level. Among the 21 studied ISSR primers, the P8A11 marker had the largest number of gene loci related to the studied physiological traits.
