other مطالعه تأثیر دوازده هفته تمرین شنای زیربیشینه بر رنگ‌پذیری Ras و Raf-1سلول‌های اپیتلیال ریۀ موش‌های صحرایی در معرض کارسینوژن NNK The effect of 12 weeks of submaximal swimming training on immunoreactivity of Ras and Raf-1 in lung epithelial cells of Wistar rats exposed to carcinogen NN پژوهشي Research <div style="border-top: none; border-right: none; border-left: none; border-image: initial; border-bottom: 5.25pt double windowtext; padding: 0cm 0cm 1pt; background: white; text-align: justify;">هدف این مطالعه، بررسی تغییرات رنگ‌پذیری  Rasو Raf-1در سلول‌های اپیتلیال ریۀ موش‌های صحرایی در معرض کارسینوژن NNK پس از تمرین شنا بود. به این منظور، 50 سر موش صحرایی به‌صورت تصادفی به پنج گروه کنترل، تمرین، تمرین-NNK، NNK و حلال تقسیم شدند. NNK به میزان 12/5 میلی‌گرم بر کیلوگرم وزن بدن، به‌صورت زیرجلدی، یک‌روز در هفته، به‌مدت دوازده هفته تزریق شد. تمرین نیز شامل دوازده هفته شنا با شدت زیربیشینه بود. درصد واکنش Ras و  Raf-1سلول‌های اپیتلیال ریه به روش ایمونوهیستوشیمی اندازه‌گیری شد. در پاسخ ایمونوهیستوشیمی Ras و  Raf-1سلول‌های اپیتلیال ریۀ گروه تمرین در مقایسه با گروه کنترل تغییر معنی‌داری مشاهده نشد (به‌ترتیب 0/889=p، 0/999=p). افزایش قابل توجهی در پاسخ ایمونوهیستوشیمی Ras و  Raf-1گروه NNK در مقایسه با گروه کنترل یافت شد (0/001=p). علاوه‌براین، در گروه تمرین- NNK در مقایسه با گروه NNK  کاهش قابل توجهی در پاسخ ایمونوهیستوشیمی Ras و Raf-1 مشاهده شد (0/001=p). به‌طورکلی، می‌توان تأیید کرد که تمرین شنا از طریق کاهش پاسخ گیرنده‌های استیل کولینی نیکوتینی و گیرنده‌های بتاآدرنرژیکی به کاهش رنگ‌پذیری Ras و Raf-1 سلول‌های اپیتلیال ریۀ موش‌های صحرایی منجر شد.</div> <div style="text-align: justify;">The aim of this study was to investigate changes in immunoreactivity of Ras and Raf-1 in lung epithelial cells of rats exposed to carcinogen NNK after swimming training. 50 Wistar rats were randomly divided into five groups: control, training, training + NNK, NNK and sham. NNK-induced groups received NNK Subcutaneously once a week at a dose of 12/5 mg per kg body weight for 12 weeks. The exercise training consisted of submaximal swimming training for 12 weeks. The percentage of reaction of Ras and Raf-1 in lung epithelial cells were measured by HIC. It was observed no significant change in the Ras and Raf-1 immunoreactivity of lung epithelial cells in training group compared to control. A significant incretion of the Ras and Raf-1 immunoreactivity was found in NNK group compared to control group (p=0.001). Furthermore, a significant reduction of the Ras and Raf-1 immunoreactivity was observed in exercise + NNK group when compared to NNK group (p=0.001).
