fa ارزیابی تمایز مورفولوژیک کاج الدار1 و دو فرم طبیعی به‌وجود آمده از آن با استفاده از الگوی الکتروفورزی صفحه SDS، صفات مورفولوژیک و برخی شاخص‌های فیزیولوژیک برای بر‌رسی و تفکیک گونهٔ کاج الدار و دو فرم مورفولوژیک به‌وجود آمده از آن، نمونه‌های برگ سوزنی نهال‌های سه ساله در شرایط یک‌سان عصاره‌‌گیری شدند. الگوی باندهای پروتئینی نمونه‌ها به روش الکتروفورز SDS-PAGE مقایسه شد. 12 صفت مورفولوژیک نهال و پایه‌های مادری سه نوع کاج نیز با یک‌دیگر مقایسه شدند. مقدار پروتئین کل نمونه‌ها به روش برادفورد و نیز محتوای کلروفیل a و b نمونه‌ها به روش هاربورن و با استفاده از اسپکتوفتومتر اندازه‌گیری شده است که الکتروفورز پروتئین تک بعدی نمونه‌ها ‌الگوهای باندی کاملاً مشابهی را برای هر سه نوع کاج بررسی شده (الداریکا و دو کاج کله‌قندی و توپی) نشان داد. میزان پروتئین کل و محتوای کلروفیل a و b نمونه‌ها با استفاده از آزمون تجزیه واریانس هیچ‌گونه تفاوت معنی‌داری را نشان نداد. در صورتی که 11 صفت از ویژگی‌های مورفولوژیک اندازه‌گیری شده در کاج الدار بیش از دو کاج دیگر بوده است. این نتایج حاکی از نبودن اختلاف بیوشیمیایی بین کاج‌ها، علی رغم وجود تفاوت‌های بارز ساختاری و مورفولوژیک است. In order to consideration and distinguishing of Mondell pine species (Pinus eldarica) and its two natural generated morphotypes, needles samples were collected from three years old seedlings in similar conditions and then extracted. Protein band patterns of the samples were studied and compared using electrophoresis method on polyacrylamide gel in the SDS-PAGE system. 12 morphological traits related to seedlings and mature individuals of the three pines, compared with each other. The amount of total protein also obtained using Bradford procedure and content of chlorophyll a and b measured by spectrophotometer with Harborne method. One dimensional electrophoresis of proteins showed perfectly similar isozyme patterns for each type (i.e. Mondell pine, Ball-shaped and Conical-shaped pines). Furthermore the amount of total protein and chlorophyll a and b contents of samples did not showed significant difference using ANOVA test. But 11 features of morphological measured traits were more in Mondell pine than the new generated pines.
